This is essential because alcohol and paraffin cannot mix directly.The tissue requires two to three xylol baths, each lasting thirty minutes. During this process, the tissue gradually becomes transparent.As the xylol replaces the alcohol in the tissue, you'll notice the sample becoming increasingly translucent. This visual change is a key indicator that the clearing process is working effectively.Throughout the clearing process, it's crucial to keep the tissue completely submerged in xylol. This ensures uniform clearing and prevents any parts from drying out.The tissue is ready for the next step when it becomes uniformly translucent. This typically takes two to three thirty-minute baths in fresh xylol.With the clearing process complete, the tissue is now ready for paraffin infiltration.The cleared tissue sample is now ready for paraffin infiltration.Inside the oven, maintained at precisely 56 to 58 degrees Celsius, we have three separate paraffin baths.The tissue undergoes three one-hour baths in liquid paraffin. This ensures complete infiltration of the tissue.After the final bath, the tissue is transferred to the embedding station, where it's placed in a mold with fresh liquid paraffin.The mold is then placed on a cooling plate, allowing the paraffin to solidify completely.The final step is proper labeling of the solidified block for accurate identification and tracking.
